Real-time PCR analysis of LOX-HPL pathway genes was performed with SYBR Green SuperReal PreMix(Tiangen,Beijing,China)according to the instruction using CFX Connect?Real-Time PCR Detection System(Bio-Rad,Hercules,CA,USA).Each reaction contained10μL 2×SuperReal PreMix Plus,1μL cDNA,0.6μL forward primer,0.6μL reverse primer and added ddH2O to 20μL.The real-time qPCR program used was 95°C for 15 min followed by 40 cycles of amplification with 95°C for 10 s,60°C for 32 s,followed by a melt cycle from 60 to 95°C.The primers used in this study were shown in Table 1.GAPDH and Ef1-αwere taken as internal controls.For all analyses,the signal obtained for target genes was normalized against the mean of cycle threshold(Ct)values of two reference genes,and relative quantification of target genes was calculated by 2–ΔΔCt method.All samples were tested in three biological replicates.
