On the basis of single-factor experiments,response surface methodology was used to investigate the effect of four factors on Y(DHA+EPA content in glycerides)after acidolysis.A Box-Behnken design(BBD)with four factors and three levels was used to generate factor combinations by using the Design-Expert 8.0.5.0(Stat-Ease Inc.,Minneapolis,MN,USA)software.The four factors were A(reaction temperature,oC),B(substrate molar ratio,capric acid/tuna oil),C(enzyme amount,based on the total substrate,%),and D(reaction time,h).The levels of the four factors based on the single-factor experiments are listed in Table 1.The experimental design with observed values is shown in Table 2.A total of 29 experimental runs was gene rated.
