《Vertical response of microbial community and degrading genes to petroleum hydrocarbon contamination in saline alkaline soil》
Soil column apparatus(inner diameter 15 cm;height 110 cm)for simulating oil contamination was shown in Fig.1.Soils were filled into column in order of sampling depth.The 700 g crude oil(provided by PetroChina Dagang Oilfield Company,see Appendix A Table S1)was added to the soil column(about5%of weight of 0–30 cm soil).The crude oil was evenly distributed in the surface soil,and stood the column for 12 hr in consideration of artificial disturbance.To avoid nitrogen and phosphorus limitations,the soils in different depth were amended with nitrogen and phosphorous fertilizer every week to obtain the C/N/P ratio of 200/15/1(m/m/m).Simulation experiment was conducted for 22 weeks period from May to October,2017.Deionized water was injected slowly into column to saturated state at initial stage,and then washed per week according to local month average rainfall as follows:
