c DNA was obtained from the total RNA of onion leaves as templates to obtain Ac FT3 gene fragment by transcriptome sequencing.According to the results of transcriptome sequencing，Ac FT3 gene clone upstream and downstream primers were designed.The full-length Ac FT3 gene was cloned and sent to Huada for sequencing.The reaction system was（20μL）:10×Taq Buffer 2μL；d NTP（2.5 mmol·L-1each）2μL；upstream primer Ac FT3-F（10μmol·L-1）0.5μL；downstream primer Ac FT3-R（10μmol·L-1）0.5μL；Tap DNA polymerase（5 U·μL-1）0.2μL；c DNA template 1μL；dd H2O 13.8μL.Reaction procedure:pre-denaturation at 94℃for 1 min；denaturation at 9℃for 1 min；52℃annealing 45 s；72℃extension 30 s；72℃extension 30 s；repeat 30 cycles；72℃10 min；storaged at 4℃.
