After horizontal centrifugation,the liquid in each centrifuge tube was layered into four layers,including the uppermost plasma layer(slightly yellow),the second membranoid lymphocyte-enriched layer(greyish white),the third colorless transparent lymphocyte separating medium layer and the lowermost erythrocyte stack layer(red).Through the comparison of the separating effects of different concentrations of Percoll solutions on sturgeon peripheral blood lymphocytes(Table 2),it was found that 70%Percoll solution(with specific gravity of 1.092 g/ml)had the optimal separating effect,with obvious lymphocyte-enriched layer interface,and the lymphocyte recovery was the highest with an average value of(89.62±8.08)%.
