To illustrate the capability and accuracy of FCS measurement on the molecular weight(MW),we used Cy3B labeled Sur A as a reference to measure the MW of Cy3B(Fig.S3a,Supporting Information).In the presence of BSA the diffusion time constant of Cy3B labeled Sur A was(352±8)?s and that of Cy3B was(85.5±0.6)?s.The theorectical MW of Cy3B labeled SurA is 47 kDa.Plugging these values into Eq.(2),MW of Cy3B was calculated to be(0.67±0.06)k Da,in good agreement with the theoretical MW of 0.658 k Da.Then,we used Cy3B labeled Sur A as the reference and measured MW of the Cy3B labeled Omp T dispersed in BSA,Tween-20,and DDM buffers(Table 1,Fig.S3).In BSA,the measured MW of Omp T was(35±9)kDa,in good agreement with the theoretical MW of single free OmpT(37 kDa),while in Tween-20 and DDM buffers the messured MW of Omp T was much larger than the theoretical MW of single free OmpT.
