To illustrate the capability and accuracy of FCS measurement on the molecular weight（MW），we used Cy3B labeled Sur A as a reference to measure the MW of Cy3B（Fig.S3a，Supporting Information）.In the presence of BSA the diffusion time constant of Cy3B labeled Sur A was（352±8）?s and that of Cy3B was（85.5±0.6）?s.The theorectical MW of Cy3B labeled SurA is 47 kDa.Plugging these values into Eq.（2），MW of Cy3B was calculated to be（0.67±0.06）k Da，in good agreement with the theoretical MW of 0.658 k Da.Then，we used Cy3B labeled Sur A as the reference and measured MW of the Cy3B labeled Omp T dispersed in BSA，Tween-20，and DDM buffers（Table 1，Fig.S3）.In BSA，the measured MW of Omp T was（35±9）kDa，in good agreement with the theoretical MW of single free OmpT（37 kDa），while in Tween-20 and DDM buffers the messured MW of Omp T was much larger than the theoretical MW of single free OmpT.
