《Isolation of Secondary Metabolites with Antimicrobial Activities from Bacillus amyloliquefaciens LWYZ003》
The inhibition zone diameter was measured by wave-crossing method
Marine Bacillus amyloliquefaciens strain LWYZ003 isolated from the sediment of Dagang Oilfi eld.Furthermore,its 16S rRNA was amplifi ed by employing two universal primers,27F(5′-AGA GTT TGA TCC TGG CTC AG-3′),1492R(5′-TAC GGY TAC CTT GTT ACG ACTT-3′).The polymerase chain reaction(PCR)program for amplifi cation was as follows:94°C for 3 min,followed by 30 cycles consisting of 94°C for 5 s,54°C for 15 s,and 72°C for 90 s,and fi nally in the extension step consisting of 72°C for 10 min.The PCR product was sequenced.The sequence was aligned with other16S rRNA sequences received from the GeneBank database and a phylogenic tree was established by Mega 5.0 with 1000bootstraps(Fig.1).The blast conclusion suggested that the16S rRNA gene of LWYZ003 strain had a 100%similarity to the 16S rRNA gene of Bacillus amyloliquefaciens BS-20(MF988364),Bacillus siamensis FJAT-hcl-15(KY653097)and Bacillus subtilis MML5328(MF688046).According to the phylogenic tree,Bacillus amyloliquefaciens is the most closely related to LWYZ003,and they also located in the same branch.Furthermore,through the experiment of molecular biology on microbial identifi cation,phenotypic identifi cation and biochemical identifi cation(Table 1),the strain was identifi ed as Bacillus amyloliquefaciens.
