As shown in Table 2,when the new buds were treated with 0.1%mercuric chloride for 3,5,and 8min,the contamination rates were 40%,25%,and15%,the death rates were 40%,45%,and 65%,and the number of new buds with callus were 2,3,and1 respectively.In this case,the disinfection effects of 0.1%mercuric chloride for new buds increased with the change of the contact time,although they were still inferior to those in the seed experiments.The maximal contamination rate was 40%,and the minimal contamination rate was 15%.However,the death rate of new buds increased drastically with the extension of the disinfection time;the peak was a65%.One possible cause might be the strong toxicity of mercuric chloride.Excessive disinfection time or improper flushing could result in the death of the explants.When the buds were disinfected with 10%sodium hypochlorite for 10,15,and 20 min,the contamination rates were 55%,30%,and 25%,and the death rates were 60%,45%,and 60%respectively.The disinfection trend of 10%sodium hypochlorite for new buds was similar to that of 0.1%mercuric chloride the extension of the disinfection time could lead to a lower contamination rate,but a higher death rate.In general,the growth status of the callus tissue was no good under all treatments,presenting a low callus rate possibly because of the improper concentrations of growth hormone and cytokinin,as well as inadequate induction time.To conclude,75%ethanol for 45 s+0.1%mercuric chloride for 5 min provided the optima disinfection effects in the bud experiments.
