During the culture of bulb explants in the dark,some of the explants began to swell and gradually developed into embryogenic callus(Fig.1 B).The calli were milky white to pale yellow and translucent.The callus induction rate in medium supplemented with different combinations of TDZ and 2,4-D was shown in Table 1.On the medium free of TDZ,the callus induction rate was 0.On the medium supplemented with 0.25 mg/L TDZ,the callus induction rate was 22.7%,and was increased to 67.3%on the medium supplemented with 0.5 mg/L TDZ.Then,with the further increase of TDZ concentration,the callus induction rate decreased gradually.On the medium supplemented with 2.0 mg/L TDZ,the callus induction rate was only 11.3%.
