With the BAC monoclonal plasmid containing Ta HKT2；2 as a template，sequencing primers were designed according to the known sequence of Ta HKT2；2，and the 5&apos；and 3&apos；terminals of Ta HKT2；2 were obtained by sequencing using ABI 3730XL DNA Analyzer.The steps above were repeated until the genomic sequences of Ta HKT2；2-B，Ta HKT2；2-D and Ae HKT2；2 were obtained，including their 5&apos；flanking sequence.With the genomic DNA and c DNA of common wheat as templates，primers were designed according to the sequencing data of the BAC monoclonal plasmid containing Ta HKT2；2 gene，and the full-length genomic sequence and c DNA sequence of Ta HKT2；2 were amplified.With the genomic DNA and c DNA of common wheat as templates，primers were designed according to the c DNA sequence of Ta H-KT2；1（Gen Bank No.U16709），and the full-length genomic sequence and c DNA sequence of Ta HKT2；1 were amplified.The used primers are shown in Table 1.
