《Molecular mapping of stripe rust resistance gene YrH9017 in wheat-Psathyrostachys huashanica introgression line H9017-14-16-5-3》
1) The segregating lines included resistant(Res)and susceptible(Sus)plants.Seg,segregating plants.2) The F2 ratios are for resistant and susceptible plants,and the F2:3 ratios are for homozygous Res,Seg and homozygous Sus lines.3) χ20.05=3.84
Among the 134 EST-STS markers,four markers showed polymorphisms in the parents and bulks.The entire F2population was genotyped with these markers.A linkage map including 10 markers(six SSR markers and four EST-STS markers)was constructed based on phenotypic and genotypic data of the F2 population(Table 2;Fig.2).These four dominant EST-STS markers,BG604577,BE471201,BE444401,and BE490440,were anchored to chromosomebin 2AL-0.85-1.00,which further confirmed the genetic linkage of the six SSR markers and four EST-STS markers with stripe rust resistance.A linkage map was constructed using software MapMaker 3.0,the result validated the association between the markers and YrH9017,where BG604577 and BE471201 were proximal to the resistance gene and Xgwm328 and Xwmc109 were distal.The closest markers to YrH9017 were BG604577 and BE471201 about1.3 and 1.8 cM in distance,respectively(Fig.3).The total genetic distance of the linkage map was 30.1 cM.
